The aim of this study was to establish, with an emphasis on cutting, concentration, time and temperature for staining, fast, efficient and reliable method for the tetrazolium test in carrot seeds. Seeds of eight commercial lots of carrots were used cultivar 'Brasilia', subject to the initial analysis for the determination of water content, germination test, first reading of germination, seedling emergence, emergence speed index, traditional accelerated aging and solution saturated NaCl (41 ° C for 48 hours). In the first study to evaluate the tetrazolium test method, after preconditioning the seeds were sectioned by two different cuts immediately immersed in tetrazolium solution (2, 3, 5 triphenylmethyl chloride) in concentrations of 0,5 and 1,0 % for 6 to 24 hours in absence of light at 30 ° C. In the second experiment, seeds remained immersed in the solution of 2, 3, 5 triphenylmethyl chloride at 0,5% and 1,0% for 6 hours at temperatures of 30, 35, 40, 45 and 50 ° C. In the third experiment the seeds were conditioned by soaking for two hours on a roll of paper under 25 ° C. Then, the seeds were sectioned and immersed in tetrazolium 0,1% solution for 1 hour under 35 ° C. This methodology was repeated two more times, with the cut changed in the first and the second cut to length beyond the staining for 2 hours. Following each method, the seeds were rinsed in distilled water and evaluated. We conclude the cut made at the time of exposing the carrot seeds in contact with the tetrazolium salt, it is the most influential factor in their coloring. The tetrazolium test can be performed in carrot seeds using two hours in pre soaking period, parallel cutting and withdrawing the embryo near the maximum tissue and immersed in 0,1% solution for two hours under 35 ° C.
