In the context of Primary Health Care (PHC), despite its strategic role in coordinating care, there are still challenges related to the systematic incorporation of safe practices as well as to the training of health professionals. Objective: To develop and validate an e-learning course on patient safety for Primary Health Care professionals. Method: An applied, methodological, and technological development research using a quantitative approach, conducted in accordance with the TIDieR recommendations and carried out in 2025. The course was developed using the ADDIE model (Analysis, Design, Development, Implementation, and Evaluation) and was organized into six self-paced modules created using Adobe Captivate® software. The course was validated using the Delphi technique by 13 judges – healthcare professionals with doctoral or master's degrees who provided care in primary healthcare – using a structured instrument based on a Likert-scale to assess the dimensions of functionality, usability, efficiency, aesthetics, content, and language. The usability assessment was conducted with 24 professionals from the Basic Health Units in the study municipality, using the same instrument that as used for the judges. To analyze the data from the two groups of participants, the Content Validity Index (CVI), Cronbach's Alpha, and McDonald's Omega were applied. Results: The course had an overall CCI of 0.95 and 0.97, respectively, for groups 1 and 2, indicating excellent validity for the dimensions assessed, with all scored above the established cutoff point (0.78), particularly in the areas of functionality, content, and language (0.97). The internal consistency indicators demonstrated adequate reliability of the overall scale (α = 0.863; ω = 0.852). The raters and professionals of Basic Health Units recognized the pedagogical adequacy, clarity of language, and applicability of the content to professional practice. Conclusion: The e-learning course proved to be a valid and promising educational technology for use in continuing education programs in the health within the Brazilian Unified Health System.
